5-Amino-1MQ (NNMT Inhibitor)
5-Amino-1MQ (5-amino-1-methylquinolinium) is a small, membrane-permeable inhibitor of nicotinamide N-methyltransferase (NNMT) studied for raising cellular NAD+ and SAM, reversing diet-induced obesity in mice, and restoring aged muscle stem cell function, and the research catalog lists it at https://onesourcepeptides.com/product/5-amino-1mq.
The target came first. Kraus et al., Nature, 2014 (PMID 24717514) found NNMT expression increased in white adipose tissue and liver of obese and diabetic mice, and showed that knocking NNMT down in those tissues protected against diet-induced obesity by raising cellular energy expenditure, with higher adipose SAM and NAD+ levels and increased polyamine flux.
The methylquinolinium inhibitors turned that target into a small-molecule tool. Neelakantan et al., Biochemical Pharmacology, 2018 (PMID 29155147) reported high membrane permeability and high selectivity, with no inhibition of related SAM-dependent methyltransferases or NAD+ salvage enzymes. In adipocytes the inhibitors lowered the NNMT product 1-MNA, raised intracellular NAD+ and SAM, and suppressed lipogenesis; in diet-induced obese mice a potent inhibitor significantly reduced body weight, white adipose mass, adipocyte size and plasma total cholesterol without changing total food intake.
Aged muscle is the second strong thread. Neelakantan et al., Biochemical Pharmacology, 2019 (PMID 30753815) injured the tibialis anterior of 24-month-old mice and found NNMT inhibition raised muscle stem cell proliferation and fusion, supported nearly 2-fold greater myofiber cross-sectional area, and increased peak torque by about 70% versus controls. Dimet-Wiley et al., Scientific Reports, 2024 (PMID 38969654) treated aged mice from 22 to 24 months and reported about 40% greater grip strength in sedentary NNMT-inhibitor mice versus sedentary controls, rising to about 60% when combined with exercise.
